مصطفى العدوي من مواليد قرية منية سمنود التابعة لمحافظة الدقهلية، أتم حفظ القرآن وهو في المرحلة الثانوية على يد مشايخ قريته، وكان قد بدأ في كتَابها، ومن ثمَّ تدرج في مراحل التعليم النظامية كأقرانه من أبناء بلدته في مدارس المحافظة حتى الثانوية، والتحق بكلية الهندسة، وبالتحديد قسم الهندسة الميكانيكية في الفترة (1397هـ - 1398 هـ)، وبعد ذلك رحل إلى مقبل بن هادي الوادعي في اليمن، وحضر دروسه بين عامي (1400 هـ - 1404هـ). ثم رجع إلى مصر، وأنشأ مسجداً صغيراً في قريته التي ولد فيها، وبدأ التدريس فيه حيث درّس صحيحي البخاري، ومسلم، ودروس في التفسير والفقه.ومع الزمن ذاع صيته فازداد عدد طلابه فأنشأ مسجداً أكبر به مكتبة كبيرة، كما أن له مؤلفات في الفقه، والحديث، ومصطلح الحديث، والتفسير، وللشيخ مشروع كبير في التفسير على صورة سؤال، وجواب، واسم هذا المشروع هو «التسهيل لتأويل التنزيل». وفي الفقه عنده كتاب «الجامع لأحكام النساء» في خمس مجلدات يشمل أربعة مجلدات للشرح، والمجلد الخامس أسئلة تطبيقية على الأربع مجلدات في المسائل المحتواة، وله كتاب آخر في الفقه بصفة عامة، وهو كتاب اسمه «الجامع العام في الفقه والأحكام»، وغيرها.
