يُعَدُّ التحقيق في علم المخطوطات أمانةً موضوعيّة ينبغي العمل على إتقائها من دون زيادة أو نقصان، أو تغيير أو تبديل في أيّ شكل من أشكال المخطوط.
فالمحافظة على المخطوط، كما هو في حالته التي يكون عليها، تعني المحافظة على تحريك الكلمات كما هي في الأصل، والإلتزام بطبع الآيات القرآنية إستناداً إلى الرّسم العثماني، وضبط الآيات القرآنيّة من خلال الإحالة إلى السّورة ورقمها، وضبط الأحاديث النبويّة وترتيبها بالأرقام لتسهل مراجعتها، ومن ثمّ إحالتها إلى مرجعها الذي استُقيت منه.
كما تُعَدُّ الأشعار المنقولة مسؤوليّةً كبرى ينبغي على المحقّق سدّ فجواتها، وخصوصاً ما يقع من التّصحيف والتّحريف في الأحرف والحركات الإعرابيّة، لذلك، فإنّ تحقيق المخطوطات لا يحتاج إلى محقّق جيّد فقط، ولكن إلى محقّق معاصر للعلوم الحديثة، يملك معلومات لغويّة وإجتماعيّة وثقافيّة.
ولأنّ جمال الخط يُعَدُّ من جمال المخطوطة، فسوف يُخصّص مبحثٌ كاملٌ حول الخطّ والخطّاطين.
