"الدر المنتظم في السر الأعظم" كتاب يتناول ما استجد عند أهل الكشف والعرفان من علامات آخر الزمان التي توصلوا إليها والتي تنبئ عن ظهور المهدي المنتظر. والكتاب يعتمد على علم الحروف والأرقام في معرفة الإشارات التي تحيط بظهور المهدي من ملاحم وفتن وغيرها. وكان لهذا العلم حظاً وافراً عند المتقدمين حتى أخذ يضمحل تدريجياً، وقد خاض فيه علماء كبار مثل: علي بن أحمد الحراني المغربي، والبسطاني، والبوني، ومحيي الدين ابن عربي، والشيخ أبي مشعر البلخي وغيرهم كثير. وعرّف هذا العلم الشيخ داود الأنطاكي بقوله: هو علم باحث عن خواص الحروف إفراداً وتركيباً، وموضوعه الحروف الهجائية، ومادته الأوقاف والتراكيب، وصورته تقسيمها كمّاً وكيفاً وتأليف الأقسام والعزائم، وما ينتج منها، وفاعله المتصرف، وغايته التصرف على وجه يحصل به المطلوب إيقاعاً وانتزاعاً، ومرتبته بعد الروحانيات والفلك والنجامة. هذا ويعدّ علم الحروف والأعداد من العلوم الشريفة، وله مبادئ عامة وحسابات دقيقة، وله أرباب وأصحاب يشتغلون به في الأزمنة الطويلة، ومنه يشتق علم الجفر والرمل.
