تبدأ القصة بزيارة من قبل دكتور مورتيمير إلى هولمز وواتسون في شارع بايكر في لندن. لدى الطبيب قصة غريبة يخبر بها عن موت صديقه وجاره العزيز، سير تشارلز باسكرفيل، وعن اللعنة الرهيبة للكلب الشرير الذي أرهب عائلة باسكرفيل لقرون عديدة. اهتم هولمز بالقصة وأرسل واتسون إلى قصر باسكرفيل ليكتشف المزيد، سرعان ما يتصل واتسون بسلسلة من الناس الغامضين. سيلدون، سجين هارب، ستابليتون، عالم الطبيعة الغريب، وأخته الجميلة، الخادم باريمور وزوجته. كلهم يتصرفون تصرفاً مثيراً للشك ولديهم كلهم أسرار مظلمة. إنهم يعيشون في عالم أراض سبخية مضببة تهب عليها الريح، في منازل لا ترحب بأحد، وأصوات مخيفة غير أرضية. في وسط كل هذا يقع قصر باسكرفيل، وسرعان ما يصبح واضحاً أن الشاب سير هنري باسكرفيل الذي وصل أخيراً وكان قد ترك القصر مع عمه سير تشارلز باسكرفيل، في خطر رهيب، هل سيتمكن واتسون من حمايته؟ أين شرلوك هولمز؟ هل ترك حقاً سير هنري ليواجه لعنة العائلة وحده؟
